Conversion of Histone to Protamine is the most important step in formation of spermatozoa. The mature sperm chromatin contains approximately 85% Protamine & 15% Histone, whereas immature chromatin contains more than 15% Histone.
In 'Aniline Blue' staining, lysine-rich Histone protein in immature nucleoprotein stains as Dark Blue in sperm head region; whereas Protamine (cysteine & arginine rich) in mature nucleoprotein stains as Eosinophilic head region.
Increased levels of persistent Histone indicate a break in the chromatin development sequence: Histone to Transition Protein to Protamine. This significantly affects the DNA packaging, is vulnerable to increased DNA Fragmentation and declined fertilization potential.
Hence, diagnosis & prognosis of male infertility can be determined by Nucleoprotein assessment.
